What's the process to build up yeast population from a slant??

I would also recommend using a turkey baster (or whatever you use to take gravity readings) to add the sterile wort to the slant, give it a good shake, and pour into the sterile starter. Repeat this again then let your first starter grow up. A stir plate would be very very helpful during this process as well.
